INTRODUCTION

The aim of the present study was to determine the optimal dose of azithromycin and doxycycline in the treatment of patients with mild to severe chronic Lyme disease (LDMD) in the context of the United States, and to evaluate the pharmacokinetic properties of these two antibiotics. This study also aimed to determine the therapeutic potential of azithromycin and doxycycline for the treatment of patients with moderate to severe LDMD in the context of the United States.

This study was conducted with a combination of laboratory techniques and validated drug bioassay for the determination of the pharmacokinetics of azithromycin and doxycycline. To avoid the influence of plasma concentrations on the pharmacokinetic parameters, all pharmacokinetic parameters for azithromycin, doxycycline, and their combination with the pharmacokinetic parameters were analyzed at the different concentrations in plasma. The bioavailability of azithromycin and doxycycline was determined using a simple dose-finding test with the dose of azithromycin and doxycycline in patients with mild to severe LDMD.

Doxycycline and the Infection Spectrum

In the last three decades, doxycycline has been the mainstay of the veterinary antimicrobial stewardship system. As a tetracycline antibiotic, it is a highly effective approach against gram-negative bacteria, and in the last two decades, it has been used for a variety of purposes, including:

  • Prescribing of doxycycline for the treatment of infections of the urinary system and gastrointestinal tract (e.g., urinary tract infections (UTIs), prostatitis, and certain types of pneumonia)
  • The use of doxycycline for the treatment of certain infections, including those associated with the following infections:

    • Acute bacterial sinusitis associated with anthrax

    • The use of doxycycline for the treatment of various infections, including:

      • Pneumocystis jirovecii pneumonia

One of the main reasons for the use of tetracyclines for a variety of bacterial infections is the potential for tetracyclines to cross the blood-brain barrier, which is a critical factor in many types of bacterial infection.

Malaria prophylaxis in travelers:Malaria prophylaxis in travelers to SSA is generally safe for travelers to SSA. However, travelers should use prophylaxis only when appropriate, regardless of travel destination. Malaria prophylaxis in travelers to SSA can be used when travel to SSA is convenient or when travel to SSA is difficult or dangerous.
